چکیده

Acrosin, a trypsin-like proteinase found in spermatozoa, is believed to play an essential role in fertilization by aiding the spermatozoon to penetrate the zona pellucida surrounding the egg (Mc Rorie and Williams, 1974). The precise cellular location of this enzyme is of considerable interest since such knowledge would aid in elucidating its mode of action. Biochemical studies have indicated that acrosin is largely bound to the sperm head, probably to the inner acrosomal membrane (Stambaugh and Buckley, 1969 ; Brown and Hartree, 1974 ; Schill and Wolff, 1974 ; Zahler and Doak, 1975), and observations of lysis of gelatin films by spermatozoa (believed to be due to acrosin

Immunocytochemical detection of acrosomal damage following cold shock: loss of acrosin from the acrosomal region of ram, bull and boar spermatozoa.

Samples of ram, bull and boar semen were subjected to cold shock and then stained using an indirect immunocytochemical method for detecting acrosin. It was found that the shock treatment abolished staining of the acrosomal region in all but a very few cells. This finding was interpreted as a great loss of proacrosin or acrosin from damaged acrosomes. Using a fluorescent label in the system, spe...

Properties of acrosomal hyaluronidase from bull spermatozoa. Evidence for its similarity to testicular hyaluronidase.

Sperm acrosomes contain hyaluronidase, the amount varying, respectively, from ram, rabbit, bull, human, boar, rat, and stallion sperm to rooster sperm that do not possess this enzyme. Hyaluronidase was partially purified from bull sperm acrosomal extracts by DEAE-cellulose chromatography. Bull sperm acrosomal hyaluronidase has an optimum pH of 3.75 and is not effected by freezing and thawing or...

A comparative study of the morphometry of sperm head components in cattle, sheep, and pigs with a computer-assisted fluorescence method

The aim of this study was to compare the sperm nuclear and acrosomal morphometry of three species of domestic artiodactyls; cattle (Bos taurus), sheep (Ovis aries), and pigs (Sus scrofa). Semen smears of twenty ejaculates from each species were fixed and labeled with a propidium iodide-Pisum sativum agglutinin (PI/PSA) combination. Digital images of the sperm nucleus, acrosome, and whole sperm ...

The occurrence of polyenoic fatty acids with greater than 22 carbon atoms in mammalian spermatozoa.

Fatty acids with carbon chain lengths greater than 22 (VLCFA) have been detected in boar, ram, bull and human spermatozoa. Saturated and mono-unsaturated fatty acids were present in all spermatozoa but, except for human spermatozoa, polyenoic fatty acids were quantitatively the most important components. Marked differences in polyenoic fatty acid composition were observed. Whereas human spermat...

Boar acrosin. II. Classification, inhibition, and specificity studies of a proteinase from sperm acrosomes.

Acrosin, a proteolytic enzyme located in the acrosome of sperm, exhibits amidase, esterase, and proteinase activity on synthetic and natural substrates containing arginyl and lysyl residues. Highly purified acrosin preparations from boar acrosomes have endopeptidase activity cleaving only the carboxyl bonds of arginine and lysine with a strong preference for arginine bonds. The Michaelis consta...
